The isolation and structure elucidation coumarin compound from bark Garcinia cymosa, provenance Kebun Raya Bukit Sari Tebo province of Jambi has been done. Powder of bark was extracted by using maceration method with methanol. The solvents fractionation being used sequencely were n-hexane, dicloromethane, and ethyl acetate. The dicloromethane fraction was coloum chromatographed by unity silica gel as stationary phase and SGP method (Step Gradient Polarity) as mobile phase. Purification fraction VI with SGP method (Step Gradient Polarity). The result of isolation is the white crystal is melt in 67-68,5oC with the amount of 0,1 gr show a single spot with some eluen on thin layer chromatography. Thin layer chromatography test and monitoring by UV 254 nm and 365 nm, fluoricence, much brighter with increase NaOH 10%, estimate of result is coumarin compound. Structure elucidation based on spectrum data UV-Vis, IR, 13C-NMR, 1H-NMR, 2D-NMR (DEPT 45, 90, and 135 MHz, HMQC, COSY and HMBC) and GC-MS, showed 2H-chromen-2-one with moleculer mass 146 and molecular formula C9H6O2.
